HDAC1 Inhibition by Melatonin Leads to Suppression of Lung Adenocarcinoma Cells via Induction of Oxidative Stress and Activation of Apoptotic Pathways

Genetic Inhibition of the Ubiquitin Ligase Rnf5 Attenuates Phenotypes Associated to F508del Cystic Fibrosis Mutation
Researchers identified RNF5 as a protein whose inhibition promoted significant deletion of phenylalanine 508 (F508del)-CFTR rescue and displayed an additive effect with the investigational drug VX-809. In addition, they found that RNF5 is differentially expressed in human bronchial epithelia from cystic fibrosis vs. control patients. [Sci Rep] Full Article

The Kinetic Response of the Proteome in A549 Cells Exposed to ZnSO4 Stress
The authors investigated the time-dependent differential expression of the proteome in A549 cells after administered with ZnSO4 for both 9 and 24 hours using 2DE. 123 differentially expressed protein spots were detected, most of which were up-regulated by Zn2+ treatment. [PLoS One] Full Article

Maresin1 Inhibits Epithelial-to-Mesenchymal Transition In Vitro and Attenuates Bleomycin Induced Lung Fibrosis In Vivo
Scientists investigated whether MaR1, a kind of pro-resolving lipid mediators, could inhibit TGF-β1 induced epithelial to mesenchymal transition in vitro and lung fibrosis in vivo. In in vitro studies, mouse type II alveolar epithelial cells were treated with different doses of MaR1 for 30 minutes and were exposed to TGF-β1 for 48 hours. [Shock] Abstract

Cigarette Smoke Affects IL-17A, IL-17F and IL-17 Receptor Expression in the Lung Tissue: Ex Vivo and In Vitro Studies
In vitro, human epithelial cell line 16HBE and A549 as well as PBMC from normal donors were stimulated with cigarette smoke extract to evaluate the IL-17A, IL-17F and IL-17R expression by flow cytometry. [Cytokine] Abstract

Synergistic Effects between mTOR Complex 1/2 and Glycolysis Inhibitors in Non-Small-Cell Lung Carcinoma Cells
By employing automated spectrophotometry, scientists found the level of glucose uptake was decreased in non-small-cell lung carcinoma (NSCLC) A549, PC-9 and SK-MES-1 cells treated with rapamycin or siRNA against Raptor, indicating that the inhibition of mammalian target of rapamycin (mTOR) complex 1 attenuated glycolytic metabolism in NSCLC cells. Resveratrol Analogue 3,4,4′-Trihydroxy-Trans-Stilbene Induces Apoptosis and Autophagy in Human Non-Small-Cell Lung Cancer Cells In Vitro
Treatment with 3,4,4′-trihydroxy-trans-stilbene concentration-dependently inhibited cell viability. It did not cause cell necrosis, but induced apoptosis accompanied by up-regulation of cleavaged PARP, caspase3/9 and Bax, and by down-regulation of Bcl-2 and surviving. [Acta Pharmacol Sin] Abstract

7-Epiclusianone, a Benzophenone Extracted from Garcinia brasiliensis (Clusiaceae), Induces Cell Cycle Arrest in G1/S Transition in A549 Cells
