TOP STORYThe authors describe the anti-cancer effects and mechanism of action of T-3775440 in small cell lung cancer (SCLC). T-3775440 inhibited proliferation of SCLC cells in vitro and retarded SCLC tumor growth in vivo. [Cancer Res] Abstract |

PUBLICATIONS(Ranked by impact factor of the journal)The authors describe a TGFβ-triggered mechanism, in which megalin, the primary mediator of alveolar protein transport, is negatively regulated by GSK3β, with protein phosphatase 1 (PP1) and nuclear inhibitor of PP1 being involved in the signaling cascade. Inhibition of GSK3β rescued transepithelial protein clearance in primary alveolar epithelial cells after TGFβ treatment. [Am J Respir Cell Mol Biol] Abstract Scientists used single-cell RNA sequencing to compare gene expression profiles between BEAS-2B lung epithelial cells chronically exposed to a sublethal dose of Cr(VI) with or without CRISPR/cas9-mediated deletion of Gene 33. [Toxicol Appl Pharmacol] Abstract Human bronchial epithelial cells were continuously exposed to 2.5 μM arsenite for about 13 weeks to induce the phenotypes of malignant transformation. They showed that Nrf2 expression was gradually decreased whereas no significant change was observed on NF-κB activation with increased time of arsenite exposure. [Toxicol Lett] Abstract Stress Fibers, Autophagy and Necrosis by Persistent Exposure to PM2.5 from Biomass Combustion Potential molecular consequences were investigated during long-term exposure of human bronchial epithelial BEAS-2B cells to fine particulate matter (PM2.5), collected from a biomass power plant. [PLoS One] Full Article Primary murine alveolar epithelial cells isolated from RAGE-/- mice revealed an altered differentiation and defective barrier formation under in vitro conditions. [PLoS One] Full Article LUNG CANCERResearchers investigated the effect of suberoylanilide hydroxamic acid on apoptosis in lung cancer cells and addressed the role of reactive oxygen species, glutathione, and thioredoxin1 levels in this process. [Mol Carcinog] Abstract The potential effect and underlying mechanisms of β-elemene on regulating endoplasmic reticulum stress to inhibit NSCLC are still unclear. A549 cells and Lewis tumor-bearing C57BL/6J mice were established to evaluate this effect. [Biomed Pharmacother] Abstract Researchers found that cisplatin induced autophagy that attenuated the sensitivity of both A549 and Lewis lung cancer cells to cisplatin. In contrast, the clinical drug andrographolide suppressed autophagy and enhanced cisplatin-mediated apoptosis in these cells. [Anticancer Drugs] Abstract |

REVIEWSExtracellular Vesicles in Lung Disease Accumulating evidence suggests that extracellular vesicles play a role in pathogenesis of lung diseases. The vesicles include exosomes, ectosomes, and apoptotic bodies. Exosomes are generated by inward budding of the membrane, subsequent forming multivesicular bodies, and releasing by exocytosis. POLICY NEWSUS Lawmakers Seek $1.1 Billion Boost for the NIH The US National Institutes of Health (NIH) would see its budget rise by US$1.1 billion in 2018, to $35.2 billion, under a spending proposal released by lawmakers in the House of Representatives. [Nature News] Editorial Marketing of Unproven Stem Cell–Based Interventions: A Call to Action Commercial promotion of unsupported therapeutic uses of stem cells is a global problem that has proven resistant to regulatory efforts. The authors suggest a coordinated approach at the national and international levels focused on engagement, harmonization, and enforcement to reduce the risks associated with direct-to-consumer marketing of unproven stem cell treatments. [Sci Transl Med] Abstract | Editorial In a Rare Move, FDA Reverses Course on Drug Developed by CEO with Ties to Trump The Food and Drug Administration has changed its tune on an experimental drug for a deadly rare disease, withdrawing a request that the company developing it run another clinical trial. The unusual move comes after President Trump met with the company’s CEO — and promised to speed up what he called a “slow and burdensome” process for drug approvals. [STAT news] Editorial
